A New Multilayered Colon-Specific Delivery
System for Prednisolone

Abstract
The purpose of the investigation was to develop a
multi-layered coated bead formulation for colon specific delivery
of prednisolone. Using an aqueous fluid-bed process, the
colon-targeting delivery system was prepared using 10% Eudragit
RS/RL (80/20), followed by 15% mixed Eudragit
RS/RL-chitosan-succinic acid layer, and finally 20% Eudragit L 100
layer. Functionality of the delivery system, as well as of each
layer, was evaluated using the USP dissolution test method. The
results show that the Eudragit RS/RL-chitosan-succinic acid mixed
layer is acting as a trigger in simulated colon fluids in this
multi-layered system.
